  Article:   QUANTIFICATION OF NICOTINE AND COTININE IN TEENAGER’S URINE.

  Abstract:   The aim of the study was to develop an Ultrahigh Performance Liquid Chromatography coupled with Mass Spectrometry (UPLC-MS/MS) method for the simultaneous identification and quantification of nicotine and its principal metabolite cotinine in teenager’s urine. Sample preparation was performed by liquid–liquid extraction followed by UPLC–MS/MS operated in electrospray positive ionization (ESI) mode with selective reaction monitoring (SRM) data acquisition. In order to measure the prevalence of tobacco consume in adolescents, 150 samples were collected and analyzed. The experiments realized for method validation reveled a linear range between LOQ (2.5 ng•mL-1) and 1000 ng•mL-1 for both nicotine and cotinine. The accuracy was less than 9 %. Repeatability and intermediate precision were ≤7.6 and ≤8.9%, respectively. Concentrations of nicotine and cotinine in smokers adolescents urine were found to range between from 2.5 to 11.170 ng•mL-1 for nicotine and 2.5 to 10.530 ng•mL-1 for cotinine, respectively. These findings significantly support the likelihood of extensive nicotine consumption through smoking teenagers in Romania.
