  Rezumat:   Selenium (Se) stimulates accumulation of carotenoids in microalgae (Zheng et al., 2017). Betaine acts as osmoprotectant for microalgae against salinity variation (Torstensson et al., 2019). The aim of our study was to determine the effects of betaine and Se on growth of Dunaliella salina in media with 3% and 7% NaCl. A high throughput assay was done in 24- and 96 well-microplates, using 2 µM sodium selenite, 0.2 µM betaine and their combination. The microalgae growth was monitored by acquiring UV-VIS spectra between 400 and 800 nm. After 14 days of cultivation, the photosynthetic pigments were extracted by dodecane and quantified spectrophotometrically (Kleinegris, 2011). Both betaine and Se caused slight growth promotion and chlorophyll accumulation. Betaine significantly enhanced carotenoids accumulation, especially in medium with 7% salt. The mixture of betaine and Se had a similar effect. Both betaine and Se act as microalgae biostimulants, because they promote microalgae growth, increase microalgae tolerance to abiotic stress, including salt stress, and enhance the accumulation of photosynthetic pigments, carotenoids and chlorophyll. Optimization of this effect is important for higher carotenoids yields from D. salina cultivated non-axenically on media with high concentrations of salt.
